Quantitative Amino Acids Analysis

Analysis of free amino acids have applications of various fields, at the beginning of this diagnosis of inherited metabolic disorders, feeding newborn with a metabolic disorder inherited comes from studies. Maple-syrup urine disease (MSUD) is an inborn error of metabolism characterized by the inherited deficiency of the branched-chain-2-oxoacid dehydrogenase complex which is required in the metabolism of the branched-chain amino acids. The detection of allo-isoleucine allows for positive identification of patients with MSUD reducing the false-positive neonatal screening results. Chromatographic separation of allo-isoleucine is readily achieved in 11 minutes analysis time with the accurate quantification using same sample.

Sample Preparation for Serum, Plasma and Urine

  1. Pipet 50 μl serum or urine sample into a centrifuge tube.
  2. Add 50 μl internal standard solution and 700 μl Reagent-1 onto sample and vortex for 10 seconds.
  3. Centrifuge at 4000 rpm for 5 minutes.
  4. Decant the supernatant into HPLC vial and inject into the system.
